It doesn't work on the collection because the speedrun mode has its own timer and each chapter must be played to get the trophy. There are skips in the game where you can skip full chapters and the trophy doesn't unlock if you do that.

Try and see. I prefer just completing hard then doing the tweak glitch on Crushing+ to get one hit kill the whole way through.

You can use tweaks only on difficulties you have beat.

In addition One hit kill unlocks after beating the game on Brutal.
