I am not sure if I just got lucky, but I have seen him spawn multiple times.  He can spawn in several places around the outside ring though, so you have to do almost a whole run through each time to try and find him.  Try going here:

When I saw this topic, I thought it was going to be about the recent change by Nintendo to make Mario's profession an athlete instead of plumber. Maybe the BL2 devs saw that shit coming. 

 

Btw, Mario is too short and fat to be anything but a plumber anyways.

The trick I always use, is to enter Ore Chasm then turn round and exit back to the Eridium Blight. By doing this, you will always spawn here when you save, quit and reload. Once you have it setup, turn left, then run over to Donkey Mong's primary spawn point and you will have an at least 70% chance of getting him to show up  ?. Whilst he's there, make sure to cut his tie off with a melee attack so you can earn some quick Badass Ranks.
