What does (1st play) mean?

Anybody knows this? Because I've started different difficulties on different save files, do they count?

My guess is gonna be either that it means the first time you do a stage in a given playthrough, which basically means when the boss is still alive (since you can re enter stages but at the end it just ends, you don't refight the boss). Or if there is a new game + that lets you carry over your stuff, then it means on a clean playthrough.

There's like a NG+ after you beat the game, all bosses in all stages will become available again. and this called 2nd play which you will not be able to get most trophies.
Newgame is always 1st play until you beat the game.

There are some trophies that require you to reach the boss with certain conditions if you kill the boss you will not be able to get them till 2nd play.

Well, this is interesting

 

In spanish the rule says "Antes de vencer a Wily" (Before beating Wily).

 

So it means, as far as I understand, you must do it before the final battle.

It means you have to do the playthrough from scratch.
