Madhouse and "Resource Manager" trophy

So I messed up my "Resource Manager" trophy for my current run. Apparently I read too late that the game forces the player to use the box 3 times in the game; before and after the "Birthday Party" segment and after the old ship escape. Now, should I just start over and only open the box during those three times or should I run the trophy during my Madhouse run?

I'd just do it on your madhouse difficulty. I know someone who did not use the item box on madhouse and did the speedrun too in the same playthrough so it's definitely possible.
