Basically, they get a load of gaming personalities to take on the role of a handler and direct Agent 47 towards his target. I imagine it'd play a bit like those bits in Watch_Dogs where you switched your view around different cameras, and directed an NPC where to go. They listed all the personalities in the video description. The last one was Jane Perry (Voice of Diana Burnwood). That's why 47 said "I look forward to working with you again, one day" to her.
